4 Principle
Extraction of a test portion in a suitable apparatus with a specified extraction solvent. Spectrometric
determination of the chlorophyll content of the extracted solution.
5 Reagent
Use only reagents of recognized analytical grade unless otherwise stated.
5.1 Extraction solvent.

Transfer to a 500 ml beaker 100 ml of anhydrous ethanol. Add to the contents of the beaker 300 ml of
anhydrous iso-octane (2,2,5-trimethylpentane) or anhydrous technical n-heptane or anhydrous petroleum
ether (essentially composed of C hydrocarbons, with a boiling range between 90 °C and 100 °C).
6 Apparatus
UsualThe usual laboratory apparatus and, in particular, are the following shall be used.
6.1 Analytical balance, capable of weighing to the nearest 0,001 g.
6.2 Mechanical grinder, blade type, or coffee mill or equivalent.
6.3 Mechanical microgrinder (see Figure 1), comprising stainless-steel tubes of approximately 50 ml
volume which can be securely stoppered, stainless-steel ball-bearings (Ø 16 mm), and an apparatus to shake

the securely stoppered tubes horizontally at a frequency of 240 min , with a horizontal displacement of
3,5 cm, or a commercial ball mill.

L'ISO (Organisation internationale de normalisation) est une fédération mondiale d’organismes nationaux

Le présent document a été élaboré par le comité technique ISO/TC 34, Produits alimentaires, sous-comité SC 2,
Graines et fruits oléagineux et tourteaux, en collaboration avec le comité technique CEN/TC 307, Oléagineux,
corps gras d’origines végétale et animale et leurs co-produits - Méthodes d’échantillonnage et d’analyse du
Comité européen de normalisation (CEN), conformément à l'Accord de coopération technique entre l'ISO et
le CEN (Accord de Vienne).
Cette quatrième édition annule et remplace la troisième édition (ISO 10519:2015), dont elle constitue une
révision mineure. Elle incorpore également l’Amendement ISO 10519:2015/Amd 1:2025.
Les modifications sont les suivantes:
— l'Annexe B, “Préparation de la courbe d’étalonnage pour déterminer le facteur k, a été ajoutée.

1 Domaine d'application
Le présent document spécifie une méthode de spectrométrie permettant de déterminer la teneur en
chlorophylle des graines de colza. Elle n'est pas applicable à la détermination de la chlorophylle dans les
huiles.

4 Principe
Extraction d’une prise d’essai dans un appareil approprié, avec un solvant d'extraction spécifié.
Détermination spectrométrique de la teneur en chlorophylle de la solution extraite.
5 Réactif
Sauf indication contraire, utiliser uniquement des réactifs de qualité analytique reconnue.
